Differential Expressions of Protamine 1 (PRM1) and Protamine 2 (PRM2) Genes as Markers of Semen Quality in Pasundan Bulls

Several gene expressions are related to the success of spermatogenesis. Protamine plays an important role in sperm DNA packaging, spermatogenesis, and quality. This study aimed isolate RNA from spermatozoa determine expression profiles protamine 1 (PRM1) 2 (PRM2). Six Pasundan bulls aged 5-8 years with a body weight 380 kg-430 kg were used for this study. The classified into group A (70%-79%) B (80%-89%) based on their motility fresh semen. In study, correlation analysis was performed between semen characteristics (volume, motility, concentration, abnormality, viability, intact plasma membrane (IPM), integrity) frozen after thawing (motility, IPM, integrity). Total isolated pellet, cDNA synthesized. Specific PCR primers transcription PRM1 PRM2 sperm. evaluated by qRT-PCR, ACTB as control. results showed that progressive positively correlated viability IPM. higher (p<0.05) than A. condition indicates existence influence parameters motility. genes could use markers quality bulls.
